Department of Medical Genetics and Fetal Medicine

The Department of Medical Genetics and Fetal Medicine belongs to one of the first in the Czech Republic. From the very beginning, it has had to rely on good co-operation with ambulatory gynecologists. Since 1977, the Department of Genetics and Fetal Medicine has been providing genetic consultations in variety of indications. In prenatal diagnostics following procedures are performed: biochemical screening of mother's blood serum (double or triple test, or more recently 1st trimester combined test), ultrasonography of fetus, in special cases fetus MR and CT, amniocentesis, chorionic villus sampling, cordocentesis and fetoscopy. Assembled samples are subsequently cytogenetically analyzed with G-banding technique, FISH or QFPCR. The department also performs DNA analyses, especially in patients with tuberous sclerosis (exclusively in Czech Republic), patients with Turner syndrome, infertile patients with microdeletions in area of AZF or as a mean of paternity determination.

Department of Medical Chemistry and Biochemistry

The Department of Medical Chemistry and Biochemistry is a modern facility of the Faculty of Medicine and Dentistry of Palacky University Olomouc with well-established tradition of research of chemical and biological features of natural compounds. The subject of research are colchicine-derived agents - alkaloids extracted from autumn crocus (Colchicum autumnale); Marian Thistle (Silybum marianum) is a source of silymarin, which is used as a liver-protecting agent. The practical outputs of the research on the effects of substances contained in the herbal extracts are food supplements. Very successful is use of sanguinarine and chelerythrine as a mean of oral hygiene and feed additives. Other fields of research include: exploration of cytotoxic and cytoprotective characteristics of substances, oxidative stress, metabolism and interaction of heterogenous agents, research on signal pathways in normal and cancer cells and cooperation in clinical studies. Department of Microbiology

Medical microbiology studies etiological agents of bacterial, fungal, viral and parasitic infections in humans. The Department of Microbiology is divided into two separately controlled, though closely connected and cooperating parts. The prevention and treatment part belongs to University Hospital Olomouc and performs mainly direct and indirect diagnostics of pathogenic micro-organisms. The second part of the department is run by the Faculty of Medicine and Dentistry, and is thus primarily pedagogically and scientifically oriented. Very close connection of scientific and clinical activities positively affects the process of education. The prevention and treatment part guarantees close connection of theoretical education with laboratory and clinical experience. The Department of Microbiology provides education for students of the following undergraduate programs: General Medicine (both in Czech and in English), Dentistry, Nursing, Midwifery, Optometry, Physiotherapy, and postgraduate study of Medical Microbiology.

Department of Anatomy

Theoretical departmentsDepartment of Anatomy is mainly focused on research of neurodegenerative Huntington´s disease (HD), as well as on disturbances of behavioural human models connected with HD field and detection of steroid hormone receptors in adult brain tissues and subsequently in embryonic tissues. The expression of androgen and oestrogen receptors, as nuclear receptors of steroid hormones, is linked e.g. with reproduction control and sexual behaviour, which are affected on HD. Newly formed laboratories cooperate with European Huntington´s Disease Network. This organisation associate and form research database from more than 16 European countries. Next research direction is connected with the anatomical significance of collateral circulation in pelvic haemorrhage, which in gynaecology and obstetrics may lead to a serious condition due to acute loss of large amount of blood, which usually lead to haemorrhagic shock with associated consequences including coagulation malfunction, impaired microcirculation and more seriously, multi-organ failure.

The tutor for incoming students is doc. MUDr. Stanislav Laichman, CSc., Head of the Department of Anatomy.

Department of Biology

Molecular methods in microbiology and routine laboratory work.¨
The aim of the project is to make student familiar with basic applications of various methods – inoculation of microorganism on agar plates, cultivation in liquid media, isolation of DNA, DNA amplification methods, HRM (high resolution melting), gel electrophoresis in routine microbiology laboratory and research. Fast and reliable characterization of the microorganisms plays an important role in adequate and effective antimicrobial therapy. The molecular methods can minimize the time of identification by conventional methods. The polymerase chain reaction is basic molecular method, when isolated DNA is added to a mixture of several solutions in order to amplify DNA. Optimization and testing of reaction?s conditions is necessary. The post PCR analysis includes performance of the gel electrophoresis and/or high-resolution melting analysis.

Molecular bases of hematological disorders
Our lab focuses on molecular bases of hematolotgical disorders, both acquired leukemia and hereditary anemia and polycythemia. We use basic as well as advenced techniques of molecular and cellular biology (PCR, Southern blot, subcloning, in vitro hematopoetic colony assays, embryonic stem cell engineering). The applicant will join the regular lab meetings. The role of the student is mainly lab work. The outcome might be a co-authorship on a poster presentation. The student will become familiar with some of the techniques mentioned above and with molecular pathophysiology of some of the diseases studied in the lab. More information: PubMed, papers by Divoky V (Affiliations: Baylor College, M.D. Anderson Cancer Center, UAB Birmingham, MCG Augusta, CCRI Wien, Palacky Univesity Olomouc)

Department of Histology and Embryology

The role of steroid receptors related to multidrug resistance (MDR) during embryogenesis.
To find correlations between expression of PPARs ( peroxisome proliferator-activated receptor) and MDR (multidrugresistant proteins) in the embryonic tissues and embryonic cell lines – to bloc expression of MDR proteins embryonic cells. Method: immunohistochemistry . The goal of this study is to deduce if there is a linkage in signal pathways between PPARs and MDR. If so, it will be possible to inhibic expression of MDR proteins by synthetic PPAR ligands in embryonall cell lines with hope that it will be applicable to cancer tissues and helpful for circumvent multidrug resistence. We will perform immunohistochemical detection of PPARs and MDR proteins to specify tissues or organs in which these proteins occur with regard to stages of prenatal development (spatiotemporal correlations). It will be a pilot study before performing FISH (to compare the occurence of both MDR proteins and PPARs and their encoding genes).

Methylation as potential reason of gene silencing detected in DNA isolated from prostate cancer cells and/or prostate cancer human tissues.
We work on finding specific way of gene silencing and on strategies to selectively turn off specific genes The aim of the study is to determine methylation status of promoter and exon 1 of the AR gene and/or ZNF185 gene in prostate cancer.. As potential reason of gene silencing detected in DNA isolated from prostate cancer cells and/or prostate cancer human tissues. Methods: Methods of cell cultures, DNA isolation from cells or tissues, bisulfite modification of DNA, PCR, gel electrophoresis analysis, Western blotting, immunoprecipitation and immunohistochemical detection.
